Anti-Tumor Mechanisms of Attenuated Salmonella Typhimurium

Oral inoculation with a nontoxic, attenuated strain (ΔcyA, Δcrp) of Salmonella enterica serovar Typhimurium transformed with a gene for human IL-2 reduced tumor burden and improved survival in a mouse model of metastatic colon cancer in both a treatment and prophylactic murine experimental metastasis model. These effects appear to be mediated by Salmonella-induced increases in circulating and hepatic NK1.1+ lymphocytes as well as a variety of induced systemic cytokines. In addition, a similar NK cell response occurs in neuroblastoma and osteosarcoma experimental tumor models. The purpose of this project is to further characterize the local and systemic immune response of S. enterica strains expressing genes for various cytokines in a variety of murine experimental tumor models. Scientists performing this research use resources of the Supercomputing Institute to plan and analyze genetic engineering aspects of this work.
