Studies on the Structure and Phosphoregulation of Cytoplasmic Dynein

This project investigates the phosphoregulation of Drosophila cytoplasmic dynein and tests the hypothesis that phosphorylation of subunits can regulate dynein targeting and function. Previous studies of dynein phosphorylation are limited, the identified sites are controversial, and no studies have been approached in a genetically tractable system where the function of sites can be tested. In collaboration with Dr. John Yates (Scripps Research Institute), these researchers are using mass spectrometry to analyze the sites of subunit phosphorylation in the Drosophila cytoplasmic dynein complex. The priority is the identification of phosphorylation sites within the IC, LIC, and LC subunits that may function in cargo attachment. Software available through MSI is used for sequence analysis and comparisons.
